Description of the perfectGene service
Based on a gene or protein sequence, which we will either use as-is or optimize for maximum gene expression, a gene is assembled from chemically synthesized oligonucleotides. The gene is then cloned into a standard vector, and a 100% correct clone is picked by sequence screening. The deliverables include:
- 1-5 µg of purified standard vector containing the synthesized gene
- Sequence chromatograms that completely cover the synthesized gene
- 100% guaranteed sequence accuracy
- Synthesis report, including a vector map
The synthesis process can be tracked via a web interface that provides detailed information on the status of a gene as well as a secure way of transferring confidential data between Entelechon and the client.

Requirements
In order to start a gene synthesis, we just need a DNA or protein sequence of the target gene. In those cases where sequence data is not available, our bioinformatics unit can provide a search and retrieval service which identifies the sequence based on an accession number, gene name, or other criteria. We can also design consensus sequences from a set of homologous genes.
Ligation product and randomized libraries
Most genes are shipped cloned into a vector. This ensures that we can select a 100% correct clone. However, in some instances customers require a free double-strand instead. In these cases, we can ship the so-called ligation product, which is the immediate result of assembling the oligos into the full-length gene. The ligation product will inevitably contain a certain degree of erroneous DNA strands, and thus needs further screening when cloned into a vector.
However, the advantage of the ligation product is that it can contain randomized nucleotide positions where two, three or all four nucleotides are randomly incorporated at a given location within the sequence. It is important to bear in mind that the overall quality of DNA libraries is limited by physical constraints. Since oligonucleotide chemistry has a limited fidelity of about 99.5% per coupling, libraries will contain mismatch mutations and deletions. Our process ensures selection of correct sequences, and therefore our error rate is usually in the range of 0.1% per nucleotide, depending on the length of the sequence and the number and kind of randomized nucleotides.
Still, at this error rate, for a gene of 800bp length, about 45% of DNA double strands will contain at least one undesired error.

Vector construction
We leverage our gene assembly technology for the efficient de novo construction of vectors according to your specifications. Entelechon’s bioinformatics will assist in the design of a vector, and QC will ensure that the client receives a fully characterized and 100% correct plasmid.

We ensure the highest possible level of confidentiality for customer data, in particular DNA and protein sequences. Please ask for a confidentiality agreement, when ordering or inquirying for custom gene synthesis.
All sequences are treated strictly confidential, and we have set up physical and digital barriers to ensure the safety of your data. Our customer web interface uses a 128bit SSL encrypted connection for maximum data security.
