Gene to Protein All Inclusive Service
We will produce purified protein from 1 litre of HEK293 or CHO cells*, including the complete workflow:
- Gene optimization with Leto
Leto 1.0 is our proprietary software for the optimization of DNA sequences for maximum expression yields. Leto features a new approach towards gene optimization, based on a genetic algorithm.
- Gene synthesis (up to 3 kbp)
Entelechon was one of the first companies fully focused on gene synthesis. Based on broad experience, Entelechon has created a robust platform for the assembly of virtually any gene sequence.
- Sequencing
- Cloning
Subcloning of the synthesised gene into our in-house expression vectors is included in the package. Entelechon offers also subcloning of the gene into a custom expression vector; for this additional service an extra fee applies.
- Expression in HEK293 or CHO cells (1 litre)
Expression of target protein in 1 litre of transiently transfected HEK293 or CHO cells. Mammalian expression systems are more suitable than lower eukaryotes or prokaryotes for providing soluble proteins that possess relevant post-translational modifications.
- Purification of protein
The target protein, secreted in the cell culture medium, will be purified via his tag or protein A column. A second purification step like SEC or IEC can be booked separately.
- Analysis and Documentation
The package includes protein analysis by SDS-PAGE and Western Blot and documentation of the purification process.
- Turn around time: Typically 6-8 weeks

For genes longer than 3 kbp and/or cloning into a custom expression vector, an extra fee applies. The time of delivery depends on the length of the ordered gene. The offer includes one purification step with affinity purification (his tag, protein A). A second purification (e.g. SEC, IEC) can be ordered separately.
